Age-related macular degeneration (AMD) is the leading cause of irreversible vision loss in elderly Americans. Although a causative agent or agents have not yet been identified, substantial evidence suggests that components of the innate and adaptive immune systems are involved in the pathogenesis of AMD. In particular, polymorphonuclear leukocytes are recruited to the choroidal vascular bed after choroidal neovascularization (CNV) secondary to AMD occurs. Several studies have demonstrated an association between polymorphonuclear leukocyte degranulation and AMD. Since the complement system is the most important innate immune system pathway in these conditions, we investigated the role of the canine complement receptor type 2 (CR2) in the pathogenesis of AMD using a naturally occurring dog model of AMD. Experimental results demonstrate that CR2-deficient dogs are protected from the development of CNV when subjected to the Freund's complete adjuvant (FCA) model of experimental autoimmune uveoretinitis.
